皖东牛CAPN1、CAST基因多态性与肉质性状相关性

摘    要:选用135头体重(493±33)kg,年龄相近皖东育肥牛,颈静脉采血,利用DNA直接测序法测定CAPN1、CAST基因多态性;试验结束屠宰试验育肥牛,取肉样分析皖东牛肉质性状,分析肉质性状与基因多态性相关性。结果表明,皖东牛CAPN1基因存在AA型和BB型2种基因型,4个单核苷酸多态性位点(SNPs),其中内含子8的C429G和G437A位置各1个突变位点,内含子9的C572T位置1个突变位点,外显子9的G458C位置1个突变位点;CAPN1基因中度多态且极显著偏离Hardy-Weinberg平衡(P<0.01)。皖东牛CAST基因有CT基因型和CC基因型,5个SNPs位点,其中内含子8的A220G、A223G和G239A位置各1个突变位点,外显子9的C369T和G375A位置各1个突变位点;CAST基因中度多态并处于Hardy-Weinberg平衡(P>0.05)。BB基因型皖东牛肌肉剪切力肉质性状显著高于AA基因型(P<0.05),CT基因型皖东牛肌肉剪切力肉质性状显著高于CC基因型(P<0.05),基因型对其他肉质性状影响差异不显著(P>0.05)。CAPN1和CAST基因多态位点与肌肉嫩度密切相关,可作为皖东牛肉质性状提升候选基因。

Polymorphism of CAPN1 and CAST genes in Wandong cattle and correlation with meat quality traits

Abstract:A total of 135 head fattening Wandong cattles were selected with similar body weights(493±33 kg) and ages. In the mid-stage, DNA polymorphism from blood collected from jugular vein of Wandong cattles was determined by direct sequencing of CAPN1 and CAST genes. At the end of the experiment, the fattening cattles were slaughtered and the meat samples were analyzed, then analyzing the correlation between meat quality traits and gene polymorphism. The research showed that there were two genotypes of AA and BB in the CAPN1 gene of Wandong cattles, and four single nucleotide polymorphism sites(SNPs) appeared, in which the C429 G and G437 A positions in intron 8 had mutation sites. A mutation site occurred at the C572 T position of 9 and a mutation site occurred at the G458 C position of exon 9. CAPN1 gene was moderately polymorphic and significantly deviated from the Hardy-Weinberg equilibrium(P<0.01). There were CT genotypes and CC genotypes in the CAST gene of Wandong cattles, and there were five SNPs, in which the positions of A220 G, A223 G and G239 A in intron 8 occur, and the positions of C369 T and G375 A in exon 9 occurred; CAST gene was moderately polymorphic and in Hardy-Weinberg equilibrium(P>0.05). The muscle shear force traits of BB genotypes were significantly higher than those of AA genotypes(P<0.05), and similarly, the CT genotypes were significantly higher than those of CC genotypes(P<0.05). There was no significant difference in the effects of genotype on other meat quality traits(P>0.05). In summary, the polymorphic loci of CAPN1 and CAST genes were closely related to muscle tenderness, and could be used as candidate genes for improving meat quality traits in Wandong cattles.
