基于线粒体控制区全序列的长江中下游鳊的遗传多样性研究

摘    要:测定了湖南岳阳、江西都昌、上海崇明3个群体42尾鳊线粒体控制区全序列940bp,发现26个变异位点和15个简约信息位点,共检出28个单倍型。转换/颠换比为45.8。在邻接树上,来自不同地点的鳊混杂分布于各分支,没有出现明显的谱系结构和地理聚群。3个群体间的遗传距离为0.004~0.005,Fst值为0~0.05,表明3个群体间没有显著遗传分化,隶属同一种群。岳阳、都昌和崇明群体的核苷酸多样性分别为0.00501、0.00414、0.00409,单倍型多样性分别为1.00000、0.93407、0.97802,其中岳阳群体的核苷酸多态性与单倍型多样性在3个群体中最丰富,建议优先保护。

Abstract:complete sequences of the mitochondrial DNA control region (940 bp) of 42 Parabramis pekinensis from Yueyang(YY),Duchang(DC) and Chongming(CM) were directly sequenced , of which 26 variable sites and 15 parsimony-informative sites were detected in 28 haplotypes. The average ratio of transitions vs. transversions was 45.8. In the neighbor-joining tree individuals from various localities were intertwined without genealogical and geographic clusters. Genetic distances among 3 populations were 0.004~0.005, and Fst value were 0~0.05, indicating that all individuals from the 3 sites belonged to one single natural population. The nucleotide diversity of Yueyang,Duchang and Chongming population were 0.00501,0.00414 and 0.00409, respectively, and their haplotypic diversity were 1.00000,0.93407 and 0.97802, respectively. As Parabramis pekinensis in Yueyang were composed of the most abundant genetic diversity in the 3 sampled sites, Yueyang should be as priority conservation areas.
