摘    要:从黑龙江大豆根部土壤中分离一株穗霉属菌株,对其诱变菌株26-13-4所产植酸酶进行研究。该植酸酶对植酸钠的适宜水解温度为45℃、适宜pH为5.0,在4、25和45℃下热稳定性好,在pH4.0~7.0时植酸酶比较稳定;低浓度的Fe2+、Mn2+、Ca2+对酶活性有促进作用,而Cu2+、Mg2+、Zn2+、Al3+以及高浓度Mn2+、Ca2+、Fe3+对酶活性有抑制作用;K+、低浓度的Fe3+对酶活没有任何影响。结果表明,该植酸酶以植酸钠为底物时的米氏常数Km为4.4×10-5mol·L-1。

Abstract:A strain of Spicaria harting was isolated form the soil near the root of soybean in Heilongjiang Province. The phytase produced by the mutant of Spicaria harting was determined. The optimal hydrolysis temperature and pH of the phytase for phytic acid were 45 ℃ and 5.0, respectively. The phytase had better heat stability at temperature 4, 25 and 45 ℃, and better stability at pH 4.0-7.0. Fe2+, Mn2+ or Ca2+ in lower content could enhance the activity of phytase, while Cu2+, Mg2+, Zn2+, Al3+ or Fe3+, Mn2+, Ca2+ in higher content could inhibit the activity of phytase. K+ and Fe3+ in lower content had no effect on the activity of phytase. The results showed that the Km of phytase was about 4.4×10-5 mol·L-1 when sodium phytate was used as substrate.
