番茄果实高质量总RNA的提取富集及RT-PCR检测

摘    要:以番茄果实为材料,比较了3种在果实中提取RNA的方法,针对番茄果实多糖的特点,利用无水乙醇和异丙醇联合沉淀的方法除去多糖,改良了Trizol法.为了检验含量比较低的miRNA,利用Licl沉淀的方法进行富集.结果表明:改良Trizol法能够有效地去除多糖,提取到的RNA 28S rRNA亮度约为18S rRNA的2倍,A260与A280比值为1.84,A260与A230比值为1.92,RNA产率为350.4 μg/g,说明利用改良Trizol法从番茄果实中提取的RNA质量好、完整性好.由此法所得的RNA可直接用于cDNA文库构建等分子生物学实验.
