摘    要:目的]确定SKS4的最佳发酵产酶条件。方法]以SKS4为试材,通过单因素试验和正交试验对SKS4的最佳发酵条件进行了优化。结果]在酵母多糖浓度为5.0%,氮源浓度为0.4%,培养基初始pH值为7.0,接种量为4.0%,装液量为40 ml/500 ml,培养温度为37℃时,蜡样芽孢杆菌SKS4产生的异甘露聚糖酶活力达2 185 U/ml。结论]建立了最佳的SKS4产酶发酵条件,获得了较高的酶活。

The Ferment Condition's Optimization of a Bacterial Strain Which Can Producing Isomerism Mannanase

Abstract:Objective] The purpose of the study was to identify the optimum conditions for produce mannanase of SKS4.Method] By means of monofactorial experiment and orthogonal optimization test,identified the optimum conditions for produce mannanase,with SKS4 as tested materials.Result] The concentration of isomerism mannanase which extracted by ourself is 5.0%,nitrogen concentration is 0.4%,the initial pH of culture medium is 7.0,inoculum concentration is 4.0%,liquid volume is 40 ml/500 ml,temperature of fermentation is 37 ℃,the enzyme activity of producing by isomerism mannanase can be reached 2 185 U/ml.Conclusion] The excellent enzyme activity can be obtained with the optimum culticivation method.
