摘    要:RS261是一株来自红海榄(Rhizophora stylosa)叶片内对辣椒疫病具有良好防病效果的生防细菌。通过55%硫酸铵沉淀获得的抗菌蛋白液对辣椒炭疽病菌等多种植物病原菌具有强烈的抑制作用,该蛋白对热稳定、对蛋白酶K不敏感,但对胃蛋白酶和紫外线较敏感,在pH6.0~7.0时抗菌活性最强;优化后的培养基配方为:马铃薯200 g/L、蛋白胨5 g/L、蔗糖20 g/L,适宜培养条件是:初始pH6.5、接种量5%、装液量为150 mL/250 mL、振荡培养72 h时,产生的抗菌蛋白最多。

Abstract:The endophytic bacterial strain RS261 isolated from the leaves of Rhizophora stylosa was an efficient biological control agent for controlling the capsicum phytophthora blight. Antagonistic crude protein extract was obtained through 55% ammonium sulphate precipitation. It was shown on PDA plates that the development of many pathogens such as Colletotrichum gloeosporioides was effectively inhibited by the antifungal protein. They had fairly good heat stability and were resistant to proteinase K, but susceptible to pepsin and UV. Their antibacterial activity was highest at pH6.0-7.0. Single factor test results indicated that the optimal medial components for producing antifungal proteins were: 200 g/L potato, 5 g/L peptone and 20 g/L sucrose. The optimal culture conditions were: initial pH 6.5, 5% inoculation volume, 150 mL/250 mL transfer ratio and 72 h culture.
