野鸭源H6N2亚型禽流感NS基因的克隆及序列分析

摘    要:根据GenBank上已发表的H6N2亚型的非结构蛋白基因(NS)序列,设计一对特异性引物,成功从本实验室分离、保存的H6N2禽流感病毒分离株A/Mallard/SanJiang/151/06(H6N2)中克隆到NS基因序列。该克隆基因全长860bp,编码NS1和NS2两种非结构蛋白,与其他H6N2亚型的NS基因进行同源性比较,同源性为71.1%~97.2%,推导的氨基酸序列同源性为62.0%~94.6%。

Molecular Cloning and Sequencing of NS Gene of H6N2 Isolated from Wild Ducks

Abstract:A pair of primers was designed, according to NS gene sequences of subtypes of avian influenza H6N2, published in Genbank and NS gene of an avian influenza virus, isolated and stored in our laborstory, was amplified with these primers.The amplifed 860bp fragment was sequenced. The nucleotide sequence and the deduced amino acid sequence of NS gene of the virus were compared with some NS sequences published.Results showed that homologies of nucleotide and amino acid sequences were 71.1% - 97.2% and 62.0% - 94.6% respectively.
