微卫星标记分析5种鹿类动物群体遗传多样性

摘    要:利用15个微卫星标记对麋鹿、黇鹿、梅花鹿、黄麂、马鹿5种鹿类动物群体DNA多态性进行测定,共检测到95个等位基因,平均等位基因数和群体有效等位基因数分别为6..3333±2.7946个和4.7089±1.8355个,平均期望杂合度为0.5790±0.1025,平均多态信息含量为0.4483±0.1050。群体间的平均遗传分化为29.4%(P〈0.001),构建5种鹿类动物群体的系统聚类图,结果基本一致,黄麂、马鹿和梅花鹿聚为一大类,黇鹿和麋鹿为一类或各自成为一类。

Analysis of Genetic Diversity of Five Deer Populations with Microsatellite Markers

Abstract:Genomes of Cervus nippon,Elaphurus davidianus,Cervus elaphus,Muntiacus reevesi and Cervus dama were scanned with 15 microsatellite markers.A total of 95 alleles were detected.Following data were obtained:average number of alleles=6.3333±2.7946;average effective number of alleles=4.7089±1.8355;average expected heterozygosity of 15 microsatellite loci of the population=0.5790±0.1050;average polymorphism information content=0.4483±0.1050;average genetic differentiation among populations Fst=29.4%(P〈0.001).Analysis by Nei's and Reynold's genetic distance and two unrooted consensus trees constructed by UPGMA method showed that 5 species were grouped into two clusters:one was Muntiacus reevesi,Cervus elaphus and Cervus nippon and another was Elaphurus davidianus and Cervus dama.
