小麦染色体2DS雌性育性位点的连锁不平衡分析

摘    要:小麦生态遗传型雌性不育系XND126的育性受两对主效基因和微效多基因联合控制。通过SSR分子标记连锁分析,在2DS上定位了一个主基因位点。为了精细定位该位点,并验证其真实性,在59个育性正常的普通小麦品种(系)与XND126组成的较小自然群体中,用2DS参考连锁图上的14对SSR研究了标记与位点间的连锁不平衡(LD)程度,发现所有标记均存在LD现象,并找到了与XND126多态性高的品种(系),作为进一步精细定位的研究材料。依据连锁分析结果,对260个品种(系)与XND126组成的较大自然群体的连锁不平衡研究发现,主效基因位点与其最近的标记Xbarc95间的LD值最大,LD衰减在大群体中同样存在,进一步表明雌性育性主基因位点的确存在于2DS。提示利用连锁分析和关联分析相结合策略进行QTI。精确定位是一种有益的尝试。

Abstract:Previous studies suggested the female fertility inheritance in wheat could be explained by two-major-genes plus polygenes mixed model. One major locus,taf1 ,was detected on 2DS by SSR linkage analysis. To fine map the major locus and confirm the linkage analysis result, the genetic diversity between XND126 and 59 varieties of common wheat were assayed with 14 SSR markers on reference map. As the results,the varieties lines highly diverse with XND126 were determined and could be used in fine mapping population construction. Interestingly, all of the 14 markers displayed linkage disequilibrium (LD) with trait locus taf1. The LD was further analyzed by a larger population of 261 varieties including the female sterile line. The maximum degree of LD was found between locus tafl and Xbarc95,the nearest marker to tafl,and LD decay was found both in the bigger and the smaller populations. These results confirmed that the tafl was harbored on chromosome 2DS in wheat. Meanwhile the analysis suggested that the combination of linkage analysis and association analysis may be an efficient tool for QTL fine mapping and under-standing the genetic mechanism of female fertility in wheat.
