合浦珠母贝含硒谷胱甘肽过氧化物酶基因特征与表达分析

摘    要:研究了合浦珠母贝含硒谷胱甘肽过氧化物酶(命名为poSe-GPx)基因结构特征和在应激状态下的表达调控机制,其cDNA全长为1271 bp,5′-非编码区(UTR)长28 bp,3′-UTR长631 bp,包括4个RNA不稳定信号结构域(ATTTA)、1个保守的硒代半胱氨酸插入序列(SECIS)和1个位于poly(A)尾巴上游22个核苷酸处的多聚腺苷信号(AATAAA).开放阅读框(ORF)为612 bp,编码由203个氨基酸组成的多肽,分子质量约23.1 ku,理论等电点为8.29.poSe-GPx第51个氨基酸为硒代半胱氨酸(Sec),由密码子TGA编码.poSe-GPx序列中存在1个保守的GPx签名序列75LGFPCNQF82、1个活性位点序列162WNFEKF167和1个糖基化位点87NCT89.利用MatGAT软件对poSe-GPx氨基酸序列和其他物种GPx序列进行同源性和相似性分析,结果表明:poSe-GPx与其他物种GPx序列具有较高的保守性,同源性为42%-55%,相似性为56%-71%.组织表达模式分析表明:poSe-GPx mRNA在消化腺、鳃、性腺、血细胞、闭壳肌、肠和外套膜中都有表达.免疫应激试验结果显示,在2和4 h时poSe-GPx基因在消化腺中的表达显著上调.

Abstract:In this study,we described the identification and characterization of a selenium dependent glutathione peroxidase gene from pearl oyster Pinctada fucata(designated as poSe-GPx).The poSe-GPx cDNA was 1271 bp and consisted of a 28 bp 5′-untranslated region(UTR),a 631 bp 3′-UTR with four cytokine RNA instability motifs(ATTTA),and a polyadenylation signal(AATAAA) with 22 bp upstream of the poly(A),and an open reading frame(ORF) of 612 bp encoding a polypeptide of 203 amino acids with an estimated molecular mass of 23.1 ku and a theoretical isoelectric point of 8.29.The poSe-GPx sequence contained a selenocysteine(Sec) residue which was encoded by the unusual stop codon,179TGA181.A GPx signature motif,75LGFPCNQF82 and an active site motif,162WNFEKF167,and putative N-glycosylation site,87NCT89 were observed in the poSe-GPx sequence.In addition,the eukaryotic selenocysteine insertion sequence(SECIS) element was also conserved in the 3′-UTR of the poSe-GPx.Homology analysis of the deduced amino acid sequence of the poSe-GPx with other known GPx sequences by MatGAT software revealed that the poSe-GPx shared 56%-71% similarity and 42%-55% identity to the other known GPx sequences.The poSe-GPx mRNA was expressed in all tested tissues,including digestive gland,gill,adductor muscle,gonad,haemocytes,intestine and mantle.Further,the immune challenge test showed that the expression level of the poSe-GPx in digestive gland had some affirmative rules and significant differences(P<0.05) at 2 and 4 h after bacterial challenge.The results suggested that the poSe-GPx could be involved in the immune responses of pearl oyster.
