小麦雌性育性与SSR分子标记的关联分析

摘    要:为了寻找与小麦雌性育性紧密关联的SSR分子标记,并估算其表型效应,选择均匀分布于小麦基因的63个SSR标记,对包括小麦雌性不育系XND126在内的261个品种(系)组成的群体进行分析,并使用STRUCTRE软件进行群体结构评估,利用TASSEL软件的GLM模型检测多态性标记与小麦雌性育性(国内法育性D.F.和国际法育性I.F.)两种表型值的关联程度.结果表明:目标群体中存在3个亚群.基因组中多态性标记关联分析初步显示,标记Xcfd36(2AS、2DS)和Xcfd88(4AL)与小麦雌性育性两种表型值相关联,两个标记对D.F.和I.F.的效应分别是0.1585,0.113和0.087,0.0596.进一步分析发现2DS和4AL连锁群上存在一些与小麦雌性育件更加紧密关联的标记,如Xwmc25(2DS)和Xwmc232(4AL),其效应分别可达到0.5833和0.2841,而2AS上未找到独有的关联标记.分析认为小麦染色体2DS和4AL各存在一个控制雌性育性的基因位点,其中2DS上的QTL被连锁分析所证实.对与表型紧密关联的标记的鉴别对于小麦雌性育性QTL发掘的重要作用进行了探讨.

Abstract:The aims of this study were to identify the SSR markers closely associated with the female fertility, and estimate their genetic effects on female fertilities in wheat. The software STRUCTRE was used to evaluate the structure of the population with 261 wheat varieties and the advanced lines, including a female sterile line XND126, based on the test of 63 SSR markers distributed evenly in wheat genome. The association was analyzed between polymorphic markers and two sets of female fertility phenotypie values (domestic fertility-D.F, and international fertility-I.F.) through the GLM model with the software TASSEL. The results showed that there were three sub-populations in the surveyed population. The candidate markers Xcfd36 (2AS, 2DS) and Xcfd88 (4AL) were associated with the phenotypes in preliminary genome scanning, and the genetic effects of the two markers on D.F. and I.F., were 0.1585, 0.113 and 0.087, 0.0596, respectively. It was also found that some markers such as Xwmc25(2DS) and Xwmc232(4AL)on the candidate markers linkage groups were more closely associated with female fertility in further analysis than those in preliminary scanning, and the genetic effects of the two markers were 0.5833 and 0.2841, respectively, whereas no significantly associated marker was found on 2AS. h is believed that two loci associated with female fertility are located on 2DS and 4AL in wheat genome, and one of the loci on 2DS is validated by QTL mapping in linkage analysis. The screening of the markers closely associated with the female fertility is important for QTLs usage of the female fertility in wheat.
