摘    要:外植体褐化是观赏凤梨等许多植物离体培养过程中的常见问题。通过研究发现,不同种类的观赏凤梨的褐化程度不一,粉叶珊瑚凤梨褐化程度较红星凤梨和黄莺歌凤梨严重;不宜选取凤梨的花序轴和花托部分作为外植体进行诱导;而减少基本培养基中的氨态氮和2,4-D浓度,初始阶段进行7 d暗培养同样有助于减少褐化的发生。不同防褐化剂处理观赏凤梨外植体的实验表明,在培养基中添加200 mg/L的抗坏血酸可以有效防止褐化产生,而不影响外植体的培养过程。

Abstract:Explant browning is a common problem during the tissue culture of Bromelia.Study was carried out on the factors resulting in browning of Bromelia in vitro culture.Results showed that the browning of Aechmea fasciata was more severe than that of Guzmania and Vriesea charlotte,and thalamus and inflorescence were not the suitable explant.Decreasing NH4 and 2,4-D,or 7 day's dark culture would reduce the browning.Results also showed that the medium supplemented with Vc(200 mg/L) could control browning of Bromelia efficiently.
