摘    要:以公农1号紫花苜蓿5~7日龄无菌苗子叶为材料,通过农杆菌介导法将轮状病毒抗原蛋白基因VP7转入紫花苜蓿子叶中,并用卡那霉素进行筛选获得抗性植株.提取转化抗性植株叶片DNA作模板,用VP7基因特异引物进行PCR扩增,琼脂糖凝胶电泳分析表明有7株扩增出了960bp的特异条带.对7株PCR阳性植株进行PCR- Southern杂交和Southern杂交分析,有4株出现阳性杂交信号,表明目的基因已经整合到紫花苜蓿基因组中,并且是1~2个拷贝.

Abstract:5~7 days old aseptic seedling of Medicago sativa cv.Gongnong No.1 was used as material.The cotyledons of Gongnong No.1 were selected as explants for genetic transformation,Antigen Gene VP7 was transformed into Medicago sativa cv.Gongnong No.1 with Agrobacterium-mediated method.And individuals of resistance were obtained in the selection medium supplemented with kanamycin.The genomic DNA in leaf of all transgenic plants was extracted as template DNA,gene VP7 was amplified with peculiar primer,and peculiar ba...
