杏叶片离体繁殖研究

摘    要:杏叶片培养可以产生不定芽,ZT与2,4-D配合能诱导出愈伤组织,其培养基为MS+ZT4.0mg/L 2,4-D 3.0mg/L,但只有在KT与NAA配合使用时才能从叶愈伤组织中分化出少量不定芽,30g/L山梨醇对叶片愈伤组织分化不定芽的作用比同浓度的蔗糖强,愈伤组织第11次继代培养中,在NAA浓度一定(5.0mg/L)的情况下,随着KT浓度的增加,新愈伤组织产生率逐步增加,且愈伤组织幼嫩,茂密,第13次继代培养时,只有MS+NAA6.0mg/L KT2.0mg/L 山梨醇30g/L及MS+NAA5.0mg/L KT4.0mg/L+蔗糖30g/L的培养基可以产生不定芽,产生率分别为6.67%和13.33%,叶片正铺与反铺对愈伤组织的诱导率均达100%,不仅正铺所产生的愈伤组织比反铺的重量大,而且只有正铺才能诱导出不定芽。

Abstract:It may regenerate adventitious buds from apricot leaf callus.Callus could be induced from leaves on Ms medium with ZT 4.0mg/L and 2,4-D3.0mg/L,But only when KT were added together with NAA,a few adventitious buds could be regenerated on MS medium,The MS media with 30g/L sorbitol could be better than 30g/L sucrose for adventitious buds regeneration from leaf callus. In 11th subculture,callus on MS with NAA regenerated rate was rising with the increasing concentration of KT,and callus were tender and denser, and in the 13th subculture,adventitious buds could be regenerated only on MS medium with 6.0mg/L NAA、2.0mg/L KT supplemented 30g/L sorbitol or MS with 5.0mg/L NAA、4.0mg/L KT supplemented 30g/L sucrose.The regenerated rate was 6.67% and 13.33% respectively.The initiated rate of callus were all up to 100% when they were put on upright or level the media.When the leaves were put upward on the media,the callus increased and adventitious buds occurred
