| 摘 要： | 为了对河北某免疫猪场发生的疑似猪伪狂犬病进行确诊,并针对猪伪狂犬病毒开展进一步的研究工作,本试验对该猪场送检的病猪进行剖检,将其脾、肺、肾及淋巴结的混合研磨液经PK15细胞接种分离,后经PCR扩增及动物回归试验,证实成功分离的病毒为伪狂犬病毒(PRV),并命名为SH151218。根据GenBank上传的PRV及其gE基因序列,设计了1对检测引物以及1对扩增gE基因的引物,并对其gE基因进行了序列分析,SH株属于GI亚型,与哈尔滨变异株同源性最高,所分离的PRV毒株为变异强毒株。此变异株的发现对PRV变异机制的研究及研制新疫苗提供了基础。

| Abstract: | The experiment was to confirme pseudorabies from a certain immuned pig farm with suspected pseudorabies in Hebei province,and to enable the further research work on the pseudorabies virus.The pig sent by the farm was dissected,and the pseudorabies virus was isolated by the PK15 cell with the mixing suspensions of the spleen,lung,kidney and lymph nodes collected from the infected pig.The pseudorabies virus was proved to be isolated successfully by PCR amplification and animal regression test,and was named as SH151208.According to PRV and gE gene sequence uploaded in the GenBank,a pair of primers for detection and a pair of primers for gE gene amplification were designed.The gE gene sequence was then analyzed,and SH strain was conformed to be GI subtype,having the highest homology with the variant strain of Harbin HeN1.The strain isolated by the lab was the variant virulent strain.The results provide a theoretical basis on PRV vaccine research and mutation mechanism. |
