摘    要:采用直接皂化方法对样品进行前处理,用乙醚和石油醚混合液(1∶1)萃取,用气相色谱、液相色谱和比色法分别进行检测。气相、液相色谱法在25~500 mg/L范围内具良好的线性关系(R20.999),比色法在0~42 mg/L范围内具良好的线性关系(R20.997)。液相、气相色谱和比色法3个加标浓度平均回收率分别为105.5%、100.1%和102.3%,检出限分别为0.1、0.4和2.1 mg/100 g,对肉松中胆固醇含量测得值分别为52.2、51.2和102 mg/100 g。3种方法均很好地满足方法学要求,其中比色法体现总固醇类化合物的含量;气相色谱法和液相色谱法专属性更强、灵敏度更高,能更准确地反应样品中胆固醇含量。

Abstract:Saponification was applied in sample pretreatment, and cholesterol was extracted by a mixed solvent of diethyl ether and petroleum ether (1:1,V/V),then analyzed by liquid chromatography(LC),gas chromatography(GC) and colorimetric method respectively. The results showed that good linearity were obtained in the range of 25~500 mg/L by using chromatogra-phy (R2>0.999) and in 0~42 mg/L by colorimetric method (R2>0.997). By using LC,GC and colorimetric method,the average recovery were 105.5%,100.1% and 102.3%,the limit of detection (LOD) were 0.1 mg/100 g,0.4 mg/100 g and 2.1 mg/100 g, and the cholesterol content in dried meat floss were 52.2 mg/100 g, 51.2 mg/100 g and 102 mg/100 g respectively. All three analytical methods were validated, among which the colorimetric method revealed the total steroid content,and LC,GC were more accurate and sensitive in cholesterol analysis than colorimetric method.
