摘    要:以草莓品种幸香叶片为外植体,研究了叶龄、暗培养、不同颜色的滤光膜、AgNO3、不同激素配比等对叶片再生不定芽的影响,建立了高效再生体系。结果表明,5周的叶龄、暗培养5 ̄6周、附加AgNO31.0 ̄4.0mg/L利于不定芽的分化;不同颜色滤光膜对草莓分化不定芽的效应不同,红膜、黄膜、绿膜能显著提高再生频率,红膜处理能滤去紫外光、蓝紫光、绿光,再生频率达最大,蓝膜对再生频率无影响,紫膜对再生频率起显著抑制作用;MS+TDZ4.0mg/L+NAA0.1mg/L暗培养6周最适于不定芽分化,再生频率达98.9%,平均再生不定芽3.8个。

Abstract:An efficient and high frequency method for shoot regeneration from leaf discs of strawberry cultivar Sachinoka (Fragaria ananassa Duch.) was established. The factors affecting adventitious shoot regeneration, such as age of leaf,dark treatment,color plastic films,AgNO3 concentrations,plant growth regulators, were studied. Results indicated that the age of leaf of 5 weeks, dark treatment from 5 weeks to 6 weeks, addition of AgNO3 1.0 to 4.0 mg/L to the medium could increase the shoot regeneration rate. Different color plastic films had great effects on the shoot regeneration of strawberry. The presence of red, yellow and green films induced the shoot regeneration rate,and red films could greatly increase the percentage of shoot regeneration to the maximum because of the absence of ultraviolet light (<400 nm), blue light (400 to 510 nm) and green light (510 to 610 nm). Blue films had no effect on the shoot regeneration, while the purple films strongly inhibited the shoot regeneration. The regeneration rate of strawberry cultivar Sachinoka reached the highest level of 98.9% with 3.8 shoots per leaf disc in the optimum MS medium supplemented with 4.0 mg/L TDZ and 0.1 mg/L NAA after dark treatment for 6 weeks.
