硫酸酯化牡蛎多糖对四氯化碳所致小鼠急性肝损伤的影响

摘    要:为研究硫酸酯化牡蛎多糖(SCGP)对四氯化碳(CCl4)所致小鼠肝损伤的影响,采用氯磺酸-吡啶法制备得到SCGP,对CCl4致急性肝损伤模型小鼠灌胃不同剂量(200、400、800 mg/kg)的SCGP,并设对照组、模型组(灌胃生理盐水)和药物组(灌胃联苯双酯,200 mg/kg),各组连续灌胃14 d,然后给模型组、药物组和SCGP组腹腔注射CCl_4致病,18 h后处死小鼠,测定各组小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)活性和肝组织匀浆中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量,观察小鼠肝组织切片形态学变化和SCGP对小鼠急性肝损伤的影响。结果表明:小鼠肝组织切片在倒置显微镜下显示,SCGP灌胃组和药物组肝细胞病变较模型组明显减少;SCGP高剂量组(800 mg/kg)能显著降低CCl4致急性肝损伤小鼠的血清ALT、AST活性和肝脏MDA含量(P0.05),显著提高小鼠肝脏的SOD活性(P0.05)。研究表明,SCGP对经腹腔注射CCl4所致的小鼠肝损伤可起到保护作用。

Hepatoprotective effects of sulfated polysaccharide from Pacific oyster Crassostrea gigas against hepatic injury induced by carbon tetrachloride (CCl4) in mice

Abstract:The mice were simultaneously given sulfated polysaccharide from Crassostrea gigas(SCGP)prepared by a chlorosulfonic acid-pyridine method at a dose of 200 mg/kg (low SCGP group),400 (mid-SCGP group),and 800 mg/kg(high SCGP group),normal saline (control group) and biphenyl dimethylester (drug group) for 14 d by gavage. The activities of glutamic-pyruvic transaminase (ALT) and glutamic-oxalacetic transaminase(AST) in serum and hepatic superoxide dismutase(SOD) activity and hepatic level of MDA were detected and hispathologic changes were observed in the mice in the groups above with acute liver injury induced by intraperitoneal injection of carbon tetrachloride(CCl4)for 18 hours to investigate the protective effect of SCGP on the mice. Results showed that the drug-induced hepatocyte lesion was significantly reduced in the mice in the SCGP group under a inverted microscope compared with the model group.The activities of ALT and AST in serum(P<0.05) were found to be re-duced significantly and MDA levels and SOD activity were enhanced in liver tissue(P<0.05) in high SCGP,resul-ting in improvement of CCl4-induced pathological tissue damage.
