马尾藻复合菌发酵液的制备及其对橡胶苗抗寒性的影响

摘    要:【目的】利用复合菌剂液态发酵制备马尾藻发酵液,探究马尾藻发酵液在橡胶幼苗上的应用及对其抗寒性的提升作用。【方法】以芽孢杆菌、里氏木霉和黑曲霉为发酵菌剂对马尾藻进行单一菌种和复合菌剂液态发酵,以复合菌发酵液开展橡胶幼苗低温胁迫试验,温度设置为15℃,分别设5种不同用量(不施用发酵液,发酵液稀释200、400、600和800倍),与30℃条件下生长的橡胶幼苗进行对比,测定马尾藻发酵液的pH、海藻酸含量、多糖提取率及橡胶幼苗的生物量、根系活力、丙二醛(MDA)、可溶性蛋白(SP)和抗氧化酶含量。【结果】马尾藻复合菌剂发酵过程中,所有处理的pH均呈先降后升的趋势,复合菌剂处理海藻酸含量略高于单一菌种。在多糖提取率方面,复合菌剂处理显著高于单一菌种,芽孢杆菌和里氏木霉二者协同处理提取率最高,为14.08%。在低温胁迫试验中,橡胶幼苗生物量均有所下降,施用马尾藻发酵液的橡胶幼苗根系活力提高,其中马尾藻发酵液稀释600倍的处理根系活力最大,为0.051 mg/(g·h)。受到低温胁迫橡胶幼苗体内的MDA含量和SP含量均有所增加,马尾藻稀释600倍处理的MDA含量增加幅度最小,SP含量提高22%。低温条件下,除马尾藻发酵液稀释200倍处理外其余处理的POD活性均有所下降,液稀释600倍处理的SOD活性在3种抗氧化酶中最高,稀释400倍处理CAT酶活性最高。【结论】复合菌剂降解效果显著优于单一菌剂,芽孢杆菌和里氏木霉菌协同发酵马尾藻效果最优,将马尾藻发酵液稀释600倍之后可有效缓解低温对橡胶幼苗的损伤作用,提高幼苗的抗逆性。

Abstract:【Objective】Sargassum fermentation liquid was prepared by liquid fermentation of compound microbial agent, so as to investigate application of Sargassum fermentation liquid on rubber seedlings and its effects on cold resistance improvement.【Method】Bacillus, Trichoderma reesei and Aspergillus niger were taken as microbial fermentation agents to conduct liquid fermentation of single strain and compound strain of Sargassum. The experimental study on low temperature stress of rubber seedlings was carried out with compound microbial fermentation broth. The temperature was set at 15℃, and 5 different dosages were set respectively(no fermentation broth, dilution of fermentation broth at 200 times, 400, 600 and 800 times). Compared with rubber seedlings grown at 30℃, pH, alginate content, polysaccharide extraction rate of Sargassum fermentation broth, and biomass, root activity, malondialdehyde(MDA), soluble protein (SP) and antioxidant enzyme content of rubber seedlings were measured.【Result】 In the fermentation process of Sargassum compound microbial agent, pH values of all treatments decreased first and then increased, and alginic acid content of compound microbial agent was slightly higher than that of single strain. In terms of polysaccharide extraction rate, the compound bacterial agent treatment group was significantly higher than the single strain treatment group and the extraction rates of Bacillus and T. reesei were the highest, 14.08%. In the experiment of low temperature stress, the biomass of rubber seedlings decreased, and the root activity of rubber seedlings treated with Sargassum fermentation broth increased. The root activity of Sargassum fermentation broth diluted at 600 times was the largest, which was 0.051 mg/(g·h). The content of MDA and SP in rubber seedlings increased under low temperature stress. The increase of MDA content in Sargassum diluted at 600 times was the smallest, and the SP content increased by 22%. Under low temperature condition, the peroxidase(POD) activity of the other treatments decreased, except for the treatment of Sargassum fermentation broth diluted at 200 times. Superoxide dismutase(SOD) activity of Sargassum fermentation broth diluted at 600 times was the highest among the 3 antioxidant enzymes, and catalase(CAT) enzyme activity was the highest when the fermentation liquid of Sargassum was diluted at 400 times.【Conclusion】 The degradation effects of compound microbial agent is significantly better than that of single bacterial agent. The synergistic fermentation of Sargassum by Bacillus and T. reesei are the best. Diluting the Sargassum fermentation broth at 600 times can effectively reduce damage of low temperature to rubber seedlings and improve their stress resistance.
