| 摘 要： | 以pEGFP-N 1质粒,通过电转染的方法转染了猪骨髓间充质干细胞,经过G 418筛选,获得了阳性细胞克隆。以转染的猪骨髓间充质干细胞与体外成熟(in v itro m aturation,IVM)的卵母细胞构建克隆胚。结果表明:通过体细胞核移植技术,猪骨髓间充质干细胞可以有效地生产转基因囊胚,并且绿色荧光蛋白可用于转基因胚胎的筛选。 |

| Abstract: | The transfected pig marrow stromal cells(MSCs) were obtained with pGFP-N1 vector containing the GFP gene and Neo gene by electroporation.The MSCs expressing GFP were detected by fluorescent microscopy and through G418 selecting and cloned culture in limiting dilution.Nuclear transferred embryos(NTEs) were reconstructed by using transfected pig bone MSCs as donors and in vitro matured oocytes as recipients.The results suggested that using somatic nuclear technology could produce transgenic blastocyst by MSCs as donors.Transgenic embryos can be selected by GFP expressed. |
