信阳水牛GHRH基因第二外显子的SNPs检测

摘    要:目的]检测信阳水牛群体GHRH基因的SNP多态性。方法]以56头信阳水牛为材料,采集血样,提取基因组DNA,以GenBank公布的牛GHRH基因(gi:194719389)序列作为参照序列,设计PCR引物,采用PCR-SSCP和DNA测序方法对该基因第二外显子的进行突变检测。结果]经SSCP分析发现,这些个体总共出现六种不同的带型。测序结果表明:位于GHRH基因的4 461bp、4 529bp、4 845bp、5 053bp和5 058bp处分别存在C→T、T→G、G→A、G→A和G→T五个单核苷酸突变。结论]表明该基因在信阳水牛群体中单核苷酸多态较为丰富。

Abstract:Objective]This study was aimed to detect the SNP of GHRH gene. Method]The Genomic DNA was taken from 56 Xinyang buffalo blood samples. PCR primers were designed according the sequence of GHRH gene (gi. 194719389). PCR-SSCP and DNA sequencing methods were used to detect the mutations. Results]The results showed that there were six different band types in different individuals by PCR-SSCP analysis. 8 SNPs were detected, located at 4461bp (C→T mutation),4 529 bp (T→G muta tion),4 845 bp (G→A mutation),5 053 bp (G→A mutation),5 058 bp (G→T mutation),respectively. Conclusion]The results indicated that GHRH gene was polymorphic in Xinyang buffalo population
