江西典型土壤条件下烟叶类胡萝卜素代谢特征研究

摘    要:为研究江西不同土壤条件下烟叶类胡萝卜素代谢的差异,在江西省抚州广昌烟区典型的旱地紫色土和水田砂壤土条件下,对烟草(Nicotiana tabacumc L cv.K326)叶片类胡萝卜素积累规律、类胡萝卜素代谢关键酶基因的表达规律进行了比较研究。RT-PCR分析表明,类胡萝卜素合成相关基因(PSY、PDS、LCYb、ZEP)及降解相关基因(CCD1)转录水平的表达在水田沙壤土烟叶中更为活跃;但LC/MS检测结果显示,旱地紫色土烟叶中类胡萝卜素积累水平较高,尤其是在烟叶成熟后期,β-胡萝卜素、叶黄素、新黄质等含量明显高于水田沙壤土烟叶。虽然烤后烟叶中,旱地紫色土和水田砂壤土烟叶类胡萝卜素降解产物总量相近,但巨豆三烯酮、二氢大马酮、香叶基丙酮等物质在旱地紫色土烟叶中含量较高,这可能是形成紫色土烟叶风格特征的主要因素之一。

A Study of Characteristics of the Metabolism of Flue-cured Tobacco Carotenoids under Typical Soil Conditions in Jiangxi Province
Abstract:To research the differences of the metabolism of flue-cured tobacco carotenoids under different soil conditions,the same tobacco variety( Nicotiana tabacum L cv.K326) was planted in typical sandy loam paddy soil and dryland purple soil-two different soil conditions in Fuzhou City of Jiangxi Province.The carote-noid contents and the expression of several key genes related to carotenoid metabolism were analyzed.The re-sults showed that the accumulation of tobacco carotenoids was relatively abundant when planted in dryland pur-ple soil,especially at the late development stages.But,the expression of the key genes related to carotenoids biosynthesis and degradation was relatively higher when planted in the sandy loam paddy soil in Guangchang County which suggested that the carotenoid metabolism was more active under that soil conditions.At the same time ,the analysis of the neutral aroma components showed that the content of the degradation products of carot-enoids were similar under two different soil conditions;while several components such as megastigmatrienone, beta-damascone and geranyl acetone were more under the dryland purple soil condition which might be the main reason why K326 showed different aroma style.
