| 摘 要： | 为证实插入了禽流感HA、NA、M1、M2、NP基因的重组杆状病毒的表达物为具有禽流感形态的病毒样颗粒,对表达产物进行电镜、Western Blot、ELISA检测。蔗糖密度梯度离心纯化表达产物,电镜观察,可见禽流感病毒样颗粒；以Western Blot方法用HA、NA、M1抗体检测表达产物和纯化产物,可见特异性条带；NA多抗为捕获抗体、HA单抗为检测抗体建立夹心ELISA方法,检测表达产物,结果为阳性。结果显示,插入了禽流感HA、NA、M1、M2、NP的重组杆状病毒能够表达禽流感病毒样颗粒。

Qualitative study of avian influenza virus - like particles |

| Abstract: | With the aim to confirm that the expression product of the recombinant baculovirus with HA, NA, M1, M2 and NP genes were virus-like particles with avian influenza morphology, the expression product was tested by electron microscopy, Western Blot and ELISA. It was purified by sucrose density gradient centrifugation and observed by electron microscope, avian influenza virus-like particles were visible. The expression product and purified product was detected by the Western Blot method with HA, NA and M1 antibodies, and specific bands were observed. A sandwich ELISA method was established with NA polyclonal antibody and HA monoclonal antibody. It was used to test the expression product and the result was positive. The conclusion suggests that recombinant baculovirus with HA, NA, M1, M2 and NP could express avian influenza virus-like particles. |
