陕西不同核桃品种指纹图谱构建

摘    要:为实现核桃品种的高效、准确鉴定,提高良种利用率,收集21个陕西常见的核桃品种,利用聚丙烯酰胺凝胶电泳与荧光毛细管电泳检测技术进行基因组多态性分析,从20对SSR引物中筛选出6对核心引物,构建不同核桃品种的SSR指纹图谱。结果表明,6对荧光引物共检测出54个等位基因位点,平均每对引物扩增9个。21个核桃品种间的遗传相似系数(GS)在0.59~0.93。进一步分析发现引物WJR265与引物WGA331结合后,再与WJR031、WJR281、WGA321、WGA032这4个引物之一任意组合即可将21个核桃品种完全区分开。用3对高效引物进行组合鉴定21份核桃材料,可以有4种不同组合。用6对引物构建供试核桃品种的分子图谱,为陕西地方核桃良种的快速鉴定和核桃指纹数据库建立提供了数据。

Abstract:In order to achieve efficient and accurate identification of walnut cultivars and improve the utilization rate,21 common walnut cultivars in Shaanxi were collected in this study.Polyacrylamide gel electrophoresis and fluorescence capillary electrophoretic techniques were used to analyze the genomic polymorphism.Finally,6 pairs of core primers were selected from 20 pairs of SSR primers,and the SSR fingerprints of different walnut cultivars were constructed.The results showed that a total of 54 alleles were detected by 6 pairs of fluorescent primers,with an average of 9 alleles per pair of primers.The genetic similarity coefficient (GS) of 21 cultivars ranged from 0.59 to 0.93.Further analysis showed that 21 cultivars were completely distinguished by combining primer WJR265 with primer WGA331 and then with one of the four primers WJR031,WJR281,WGA321 or WGA032.Six pairs of primers were used to construct the molecular map of the tested walnut cultivars,which provided data for the rapid identification of local walnut cultivars in Shaanxi and the construction of walnut fingerprint database.
