赤楠叶总黄酮微波-碱水法提取工艺及其含量动态研究

摘    要:目的]探索赤楠叶总黄酮的提取工艺及含量动态变化。方法]采取微波-碱水法提取赤楠叶总黄酮,通过单因素试验与正交试验对其工艺条件进行正交优选,以优选工艺条件提取不同月份的赤楠叶总黄酮,紫外分光光度法测定与考察其含量动态变化。结果]微波功率对赤楠叶总黄酮提取率有显著影响,碱水pH值有极显著影响,优化的微波-碱水法提取工艺条件为:以原料50倍量的pH值为12的碱水,在微波功率300 W的条件下提取8 min,提取次数为1次;赤楠叶总黄酮含量在11月份左右达到最高,8月份左右降到最低。结论]微波-碱水法提取工艺是一种高效的、节约成本的赤楠叶黄酮提取方法,叶黄酮提取的最佳采叶期以11~12月为宜。

Abstract:Objective] The study aimed to discover the extraction technics and dynamic content of total flavonoids in Syzyginm buxifolium leaves.Method] Total flavonoids of Syzyginm buxifolium leaves was extracted by the microwave-alkali water method.And its optimum conditions were found by the single factors experiments and orthogonal test,and the change trend of content in one year was found by ultraviolet spectrophotometry.Result] Microwave power had significant effect on the extraction rate of total flavonoids,and Alkali water pH value had more significant effect.The optimum extraction conditions of microwave-alkali water method were as follows: The ratio of material to liquid was1∶50,the alkali water pH value was 12,microwave power was 300 W,microwave treatment time was 8 minutes,extracting times was 1 times.Around November,the leaf flavone content reached the peak,but around August it reached the lowest point.Conclusion]The microwave-alkali water extracting technics was the more effective and economical method to extract Syzyginm buxifolium leaf flavonoids,and the period of best-collecting-leaf was from November to December.
