| 摘 要： | 为探讨PRRSV非结构蛋白4(NSP4)功能,体外构建了pEGFP-N1-nsp4真核表达载体,转染Marc-145细胞,利用NSP4-GFP融合蛋白携带的绿色荧光标记,研究NSP4蛋白在Marc-145细胞中的定位,并根据蛋白疏水性设计构建含有nsp4基因N端不同长度的重组pEGFP-N1表达载体,转染细胞以进一步了解NSP4蛋白是否存在核定位信号序列及其在细胞中的分布。试验结果表明:融合蛋白主要位于细胞核,部分在胞浆内。PRRSV NSP4蛋白铰链区(hinge region,HR)145～168氨基酸是决定蛋白核内定位的功能区,该片段缺失影响NSP4蛋白进入细胞核。结论:体外转染细胞内表达的NSP4蛋白主要分布在细胞核内,HR区是其核定位信号,NSP4蛋白通过HR区信号作用主动进入细胞核内。
