摘    要:目的]探究利用秋水仙素诱导紫穗槐多倍体的优化条件。方法]以紫穗槐成熟种子为试材,采用正交试验,研究了不同秋水仙素浓度、处理根长(≤2、2~4mm)、支撑物和光照条件对秋水仙素诱导紫穗槐多倍体的影响。结果]秋水仙素浓度是影响紫穗槐染色体加倍的主要原因,0.03%~0.09%处理浓度均有一定诱变效果,诱导率可达57%~76%。其次是处理根长,处理2~4mm的根得到的膨大的根尖比处理〉4mm根的情况下数量多。3种支撑物中以纱布效果最佳,棉花与纱布的效果较为接近。3种光照中,半光条件效果最佳。结论]利用秋水仙素诱导紫穗槐多倍体的优化条件是:根长2~4mm的紫穗槐根尖用浓度0.03%的秋水仙素诱导,以棉花或纱布为支撑物,全光或半光条件下48h。

A Preliminary Inquiry on Optimal Conditions for Inducing Polyploidy of Amorpha fruticosa

Abstract:Objective]The study aimed to explore the optimal conditions for inducing polyploidy of Amorpha fruticosa by colchicine.Method]With the mature seeds of A.fruticosa L.as the tested material,the orthogonal experiment was used to research the effects of colchicines on inducing polyploid of A.fruticosa under different conditions of different colchicines concn.,root length,supporting materials and light.Result]The colchicine concn.was the main reason for inducing polyploid of A.fruticosa and its treating concn.of 0.03%-0.09% all had some mutagenesis effect,with the induction rate of 57%-76%.Te treated root length with 2-4 mm could get more expanding root tips than the one with > 4 mm.Among 3 kinds of supporting materials the Absorbent gauze got the optimum effect and the effect of cotton was more close to that of the Absorbent gauze.Among 3 kinds of light,the half illumination of light got the optimum effect.Conclusion]The optimal conditions for inducing polyploidy of A.fruticosa by colchicine was as following:with 0.03% of colchicines,root length of 2-4 mm,supporting material of cotton or Absorbent gauze,and full or half illumination of light under 48 h treatment.
