| 摘 要： | 根据禽多杀性巴氏杆菌psl基因序列,设计了1对引物,从猪源多杀性巴氏杆菌5:A型Ts-8株中扩增出453bp的psl基因,将其克隆到pMD18-T载体后测序。序列分析表明,该基因与禽多杀性巴氏杆菌T16株的psl基因序列只相差2个碱基,同源性高达99%;与流感嗜血杆菌编码P6蛋白的基因序列同源性为83%。猪psl基因的推导氨基酸序列与禽psl基因、P6蛋白的氨基酸序列同源性分别为100%,87%。结果表明,多杀性巴氏杆菌psl基因在猪和禽的菌株中是高度保守的,而且与P6蛋白的氨基酸序列具有较高的同源性。

| Abstract: | A pair of primers were designed based on the sequence of psl gene of fowl cholera(U16849),which could amplify a 453bp PCR segment from the template of Pasteurella multocida serotype 5:A strain Ts-8.The amplified DNA fragment was cloned into pMD18-T and identified by sequencing.Analysis of sequence showed that the nucleotide sequence of the cloned gene had only two different bases from fowl cholera strain T16(99% homology) and 83% homology with the gene encoding the P6 protein of H.influenzae.The deduced amino acid sequence of porcine psl gene had 100% homology with the deduced amino acid sequence of the psl gene of fowl cholera strain T16 and 87% homology with P6 protein.The results showed that psl gene is highly conserved in the porcine and avian strains. |
