贵州李SSR反应体系的优化

摘    要:为了得到贵州李SSR扩增最优化反应体系,以贵州李品种早酥为试材,采用单因素试验的方法,对SSR反应体系的主要成分及退火温度进行优化。结果表明:在25μL反应体系中,模板DNA用量为30ng,引物浓度为0.80μmol/L,Master Mix用量为12.5μL;退火温度在46~50℃均能得到清晰的条带。利用此反应体系对16个贵州李品种进行PCR扩增及电泳检测,扩增结果清晰且不同品种间的DNA谱带多态性丰富。表明,该体系适合贵州李的亲缘关系分析。

Abstract:In order to get optimized SSR amplification reaction system of Guizhou plum,the Guizhou Zaosu plum was used to optimize the main components and annealing temperature of SSR reaction system through single factor experiment.The results showed that the template DNA dosage was 30 ng.The primer concentration was 0.8 μmol/L primer and the Master Mix dosage was 12.5 μL in the 25 μL reaction system.The clear ladder could be got when the annealing temperature was between 46~50℃.Through the above reaction system,the PCR amplification and electrophoresis detection of 16 cultivars of Guizhou plum were conducted.The amplification results were clear and the polymorphism of DNA bands was abundant.All of these indicated that this system was suitable for analysis of phylogenetic relationship of Guizhou plum.
