禽巴氏杆菌C48-1外膜蛋白H基因克隆及序列分析

摘    要:根据已发表的外膜蛋白基因核苷酸序列设计一对特异性引物,应用PCR方法扩增出禽巴氏杆菌5:A C48-1株的ompH全长片段,将ompH进行T—A克隆、序列测定和分析。结果表明,ompH全长1597bp,含有一个1056bp的开放性阅读框架(ORF),编码352个氨基酸,前20个氨基酸组成信号肽。与已知的15个血清型及痘苗株CU的ompH的核苷酸及其推导的氨基酸序列比较,核苷酸同源性在51.5%~98.7%之间,氨基酸同源性在39.3%~98,7%之间。氨基酸多序列比较显示,血清型特异性抗原表位位于60~80位和200~220位氨基酸处。

Abstract:According to ompH of pasteurella multocida published in GenBank, a pair of primers were designed. Using the primers, full-length ompH of C48-1 was amplified by PCR. It was cloned into pMD18-T vector and then sequenced. The result showed that the full-length ompH composed 1 597 bp.The coding region of ompH is 1056 bp at length. The predicted primary protein is composed of 352 amino acid with a 20-amino-acid signal peptide, After comparing with ompH from 15 pasteurella multocida serotype as well as the CU vaccine strain, it showed that nucleotide sequence homologies were 51.5 % to 98,7 % and the deduced amino acid homologies were 39,3% to 98.7%. By amino acid sequence alignment of ompH, two hypervariable regions-amino acids 60 to 80 and 200 to 220 which contain serotype specific epitomes and may induce protective antibodies were identified.
