韩国型鸭肝炎病毒福建株的分离与鉴定

摘    要:从福建省某养鸭场罹患鸭肝炎的病死鸭中分离到1株大小35~45 nm、十二面体对称的无囊膜病毒(暂命名为FJ01株),其鸭胚半数致死量(ELD50)为10~(-2.69)/0.2 mL,该病毒可被特异性韩国型鸭肝炎病毒免疫血清所中和,可被韩国型鸭肝炎病毒特异引物所扩增,而不能被1型鸭肝炎病毒引物所扩增。遗传进化分析表明鸭肝炎病毒FJ01株与韩国型鸭肝炎病毒关系密切,它们在进化树中共处一群。

Abstract:A virus strain, designated as FJO1, was isolated from a dead duckling characterized by duck hepatitis in Fujian province. Virus particles were non-enveloped, icosahedral symmetry, and with a diameter of 35~45 nm. The 50% embryo lethal dose (ELD50) in FJO1 was determined to be 10~(-2.69)/0. 2 mL with 10-day-old duck embryos. The virus could be neutralized by serum against South Korea-type of duck hepatitis virus (DHV). A pair of primers to South Korea-type DHV could be used to amplify the specific fragment from the genome of DHV FJO1 by RT-PCR, but the specific primers to DHV type 1 could not. Phylogenetic analysis showed that DHV FJO1 was close to South Korea-type DHV and they could be subdivided into a same cluster.
