菜籽粕脱毒及菜籽蛋白分离的工艺研究

摘    要:目的]研究双低菜籽粕的蛋白提取工艺,为菜籽粕的开发利用提供理论指导。方法]以脱毒后的双低菜籽粕为原料,采用单因素试验研究不同因素对菜籽蛋白提取率的影响,并采用响应面法对影响因素进行优化。结果]单因素试验结果表明,菜籽粕蛋白最佳提取条件为:pH值13,温度40℃,料液比1∶12,提取时间40 min,提取次数3次。在此基础上用响应面法对pH值、提取温度和料液比的最佳水平做进一步研究和探讨,通过对二次多项回归方程求解得知,在自变量分别为pH值12.9、温度40.8℃、料液比为1∶12.6时,双低菜籽粕蛋白提取率最大预测值为57.73%。结论]丙酮-乙醇溶液对菜籽粕具有一定脱毒效果,响应面法所得数学模型在预测各因素对菜籽蛋白提取率的影响上具有良好的指导性。

Research on the Technology of the Detoxification of Rapeseed Meal and the Isolation of Its Protein

Abstract:Objective] The theoretic guidance for the development and utilization of rapeseed meal was provided through the research on the extracting technology of protein from rapeseed meal.Method] The double-low rapeseed meal detoxified being taken as raw material,the effect of the different factors on the extracting rate of protein was experimented and the affecting factor was optimized with response surface method.Result] The single-factor experimental result indicated that the best extraction technology of the protein from rapeseed meal was the pH value was 13;the temperature,40 ℃;the ratio of material to liquid(g/ml),1:12 and the extraction time,40 minutes for 3 times' extraction.Based on the result,the level of the pH value,the extraction temperature and the ratio of material to liquid was further optimized with response surface method and while the argument was the pH value of 12.9,the temperature of 40.8 ℃ and the ratio of material to liquid of 1:12.6,which was produced by means of the quadratic regression equation,the optimum prediction yield of the protein from rapeseed meal was 57.73%.Conclusion] The rapeseed meal could certainly be detoxified in the treatment of acetone-ethanol solution.The guidance of the mathematical model,formed through response surface method,for the prediction of all factors affecting the extracting efficiency of the protein from rapeseed meal was feasible.
