共轭亚油酸高产菌株的筛选及培养基的优化

摘    要:目的]从自然发酵的酸菜汁中筛选共轭亚油酸高产菌株以及优化该菌株的培养基。方法]以3种自然发酵的泡菜为研究对象,通过溴甲酚紫平板筛选产酸细菌,革兰氏染色法初步鉴定,紫外吸收光谱法检测发酵液中共轭亚油酸含量等方法,从酸菜汁中筛选到一株共轭亚油酸高产菌株QL2,对其培养基成分碳源、氮源和无机盐离子及底物亚油酸的添加量进行优化。结果]菌株QL2共轭亚油酸产量达23.263μg/ml,亚油酸转化率为3.88%。优化后的培养基组成为:乳糖20 g/L,酵母膏30 g/L,CH3COONa2 g/L,MgSO4.7H2O0.8 g/L,K2HPO4.3H2O3 g/L。优化培养基组成成分后,底物亚油酸添加量为0.40%(V/V)时,共轭亚油酸产量达288.740μg/ml,转化率高达7.21%。结论]研究为产共轭亚油酸菌株的筛选及培养基的优化提供了参考。
