摘    要:目的]建立柱型苹果鲁加16号的最佳叶片再生体系。方法]以柱型苹果鲁加16号的3年生试管苗叶片为外植体,研究基本培养基、激素种类及浓度配比、暗培养、接种方式及外植体苗龄对鲁加16号叶片再生的影响。结果]3种培养基中以MS培养基为最好,鲁加16号叶片在MS培养基上的再生频率和平均生芽数分别为62.7%和3.3个,明显高于B5(40.2%和2.1个)和1/2MS(50.4%和2.6个)培养基。该叶片再生芽数与激素的浓度及其配比密切相关。该叶片最适宜的培养基是:MS+6-BA4.0 mg/L+NAA0.10 mg/L+蔗糖30 g/L+琼脂7 g/L。叶片的正反面接种方式对叶片再生没有显著影响。暗培养对叶片再生不是必需的,但暗培养2周左右能明显提高叶片的再生能力。结论]选择苗龄25~30 d、叶色嫩绿、长势较强的叶片作为外植体,可以显著提高苹果离体叶片的再生频率。

Abstract:Objective] The purpose of study was to extablish the optimum leaf regeneration system of columnar apple Lujia 16.Method] With the leaves of 3-year-old tube seedlings of columnar apple Lujia 16 as explants,the effects of basic medium,hormone kindes and concn.matching,dark culture,inoculation modes and explant age on the leaf regeneration of columnar apple Lujia 16 were stduied.Result] Among 3 mediums MS was best.The regeneration frequency and the mean bud number of Lujia 16 leaves on MS medium were 62.7% and 3.3 resp.,being obviously higher than on B5(40.2% and 2.1) and 1/2MS(50.4% and 2.6) mediums.The regeneration bud number had close conrelation to the hormone concn.and their matching.The optimum medium for Lujia 16 leaves was MS 6-BA 4.0 mg/L NAA 0.10 mg/L sucrose 30 mg/L agar 7mg/L.The inoculation modes of the front and the back of leaves had no significant effect on the leaf regeneration.Although dark culture was not necessary for leaf regenerating,dark culture for about 14 days could obviously enhance the regeneration ability of the leaves.Conclusion] Selecting 25-30 d old healthy leaves as explants could obviously enhance the regeneration frequency of apple leaves in vitro.
