北海文昌鱼遗传多样性的RAPD分析

摘    要:【目的】了解北海文昌鱼的遗传背景,探讨其遗传多样性发生变化的原因并提出保护建议。【方法】运用RAPD技术对33份北海文昌鱼样本进行遗传多样性检测。【结果】从30个随机引物中筛选出8个重复性好、条带清晰的引物;对每个样品的基因组DNA进行扩增,共获得56个RAPD位点,其中多态位点28个(占50.00%),RAPD产物分子量在220~1500bp之间;个体间遗传相似系数平均为0.8736,个体间遗传距离平均为0.1264;群体Shannon信息指数为0.2190,Nei's基因多样性指数为0.1391;中性检测结果表明所有位点均符合中性假说。【结论】相对于厦门文昌鱼和青岛文昌鱼,北海文昌鱼遗传多样性较低,但仍具有一定的遗传多样性,今后应进一步加强北海文昌鱼种质资源的管理和保护。

Abstract:Objective]The present study was aimed to study the genetic background of Beihai amphioxus(Branchiostoma belcheri Gray) and explore the cause of genetic diversity amongst them.Method]RAPD markers were used to assess the genetic diversity amongst 33 samples of Beihai amphioxus.Result]Of 30 random primers used for the amplification of amphioxus genomic DNA,8 produced reproducible and distinct bands varying in size from 220 to 1500 bp.Fifty-six loci were detected,28 of which(50.00%) were polymorphic.The average genetic similarity index and average genetic distance were 0.8736 and 0.1264,respectively.The Shannon index and Nei's gene diversity were 0.2190 and 0.1391,respectively.The results of neutral detection showed that all loci were in accordance with the neutral hypothesis.Conclusion]As compared to amphioxus(Acipenser sinensis,Barracuda,Lateolabrax sp) in Xiamen and Qingdao in China,Beihai amphioxus showed relatively lower genetic diversity.The suggestions were made for management and protection of Beihai amphioxus germplasm resources in future.
