地黄萎蔫病致病菌的鉴定及防治药剂筛选

摘    要: 从河南省温县地黄种植园采集具有典型萎蔫症状的地黄病株,分离、纯化出5株病原菌。对其进行形态学观察、致病性检测及分子生物学分析,其中1株鉴定为黄瓜织球壳菌(Plectosphaerella cucumerina),为地黄的新型致病菌。将该病原菌引起的地黄病害暂命名为地黄织球壳菌萎蔫病。该病原菌对光照敏感,最适培养基为PDA培养基;菌丝最适生长温度为25 ℃、最适pH值为8,最适碳氮源分别为可溶性淀粉和硝酸钠。室内防治药剂筛选结果表明,选用的11种杀菌剂中,有6种对P. cucumerina表现出不同程度的抑制作用,其中500 g·L-1苯醚甲环唑·丙环唑乳油对病原菌菌丝生长的抑制效果最好,EC50为0.255 3 mg·L-1,可作为防治地黄织球壳萎蔫病的优选药剂。本研究为地黄萎蔫病的识别与防治提供了科学依据。

Abstract:Rehmannia glutinosa plants with typical wilting symptoms were collected from R. glutinosa plantation in Wenxian County, Henan Province. A total of five pathogens were isolated and purified. Based on the morphological observation, pathogenicity detection and molecular biological analysis of the pathogens, one of the pathogens was identified as Plectosphaerella cucumerina, a new pathogen of R. glutinosa. The disease caused by P. cucumerina was named the Plectosphaerella Rehmannia Wilt. The study of biological characteristics showed that P. cucumerina was sensitive to light, and PDA medium was the most suitable medium for the growth of P. cucumerina. The optimal temperature for mycelium growth was 25 ℃, the optimal pH value was 8.0, and the most suitable carbon and nitrogen sources were soluble starch and sodium nitrate, respectively. In addition, the bioassays were carried out and the results showed that six of the eleven selected fungicides exhibited different degree of inhibitory effects on P. cucumerina. Among these 500 g·L-1 Difenoconazole and Propicondzole EC had the best inhibitory effect on mycelial growth, EC50 was 0.255 3 mg·L-1. It can be used as the optimal fungicide to control the Plectosphaerella Rehmannia Wilt. The results presented herein provided a scientific basis for the identification and management of the Plectosphaerella Rehmannia Wilt caused by P. cucumerina.
