摘    要:以广西花生主栽品种桂花17和桂花22的成熟种子胚小叶为外植体,探讨外植体处理方式、不同激素组合和蔗糖浓度对外植体不定芽诱导、继代、生根培养的影响。结果表明,在MS培养基中预培养0、4d的花生种子的胚小叶利于不定芽的诱导,具有较高的分化率;近叶柄1/3处横切的小叶不定芽分化率较远离叶柄部位的高。适宜小叶不定芽分化的蔗糖浓度为40~50g/L,40、50g/L蔗糖分别利于桂花22、桂花17不定芽的分化。培养基MS+4.0mg/L 6-BA+1.0mg/L NAA+2.0mg/L AgNO3对桂花17不定芽的分化效果较佳,MS+6.0mg/L 6-BA+1.0mg/L NAA+2.0mg/L AgNO3利于桂花22不定芽的分化,两者的不定芽分化率均达100%;继代培养基MS+1.5mg/L 6-BA+0.4mg/L NAA有利于促进两个花生品种的不定芽成苗,而最佳生根培养基为1/2 MS+0.5mg/L 6-BA+2.0mg/L NAA。已构建的花生植株再生体系,不定芽分化率高,再生植株多,适合进行花生的遗传转化。
