猪源枯草芽孢杆菌BL-Y9益生特性研究及小鼠安全试验

摘    要:本研究对实验室自行分离保存的猪源枯草芽孢杆菌BL-Y9进行抑菌、产酶、体外消化率等益生特性研究,并测定了其对小鼠的安全性。结果表明:菌株BL-Y9对猪大肠杆菌K88、鸡大肠杆菌O78、鸡白痢沙门氏菌CVCC533及金黄色葡萄球菌C56011有较好的抑菌效果,抑菌圈直径均可达到10 mm以上,发酵上清液中性蛋白酶及纤维素酶活性分别可达184.94、9.68 U/mL|经模拟动物体消化道的试验结果表明:菌株BL-Y9对三种不同饲料组合的消化效果较好,其中以对玉米面+豆粕组合消化效果最好,经作用后,消化液粗纤维消化率、游离氨基酸增加幅度、可溶性氨基酸增加幅度、还原糖增加幅度及酸溶蛋白增加幅度分别为37.79%、152.94%、438.11%、325.62%、131.39%。动物试验结果显示,每只小鼠灌胃1 ~ 100亿剂量菌体是安全的。菌株BL-Y9可作为畜禽饲料添加剂使用。 关键词] 枯草芽孢杆菌|产酶特性|抑菌特性|消化率|小鼠安全性

Abstract:The probiotic characteristics such as bacteriostasis,enzyme production and in vitro digestibility of Bacillus subtilis BL-Y9 were studied,and the safety of mice was determined.The results showed that the strain BL-Y9 had good antibacterial effect on Escherichia coli K88,Escherichia coli O78,Salmonella gallinarum CVCC533 and Staphylococcus aureus C56011,and the diameter of the inhibition zone could reach more than 10 mm.The neutral protease and cellulase activity of the fermentation supernatant were 184.94,9.68 U/mL;The results of the simulated animal digestive tract showed that the strain BL-Y9 had better digestion effect on the three different feed combinations,and the best effect on the combination of cornmeal and soybean meal was observed.Afterwards,the digestibility of crude fiber,free amino acids,soluble amino acids,reducing sugar and acid-soluble protein were increased 37.79%,152.94%,438.11%,325.62%and 131.39%,respectively.Animal experiments showed that each mouse was safe in the range of 100 million~10 billion doses of bacteria.Based on the results mentioned above,the strain BL-Y9 could be used as a feed additive for livestock and poultry.
