| 摘 要： | 利用快速扩增cDNA末端(rapid amplification cDNA end, RACE)技术获得草鱼细胞凋亡抑制蛋白基因全长cDNA序列为2 093 bp,含有1个1 944 bp的开放阅读框,5′非编码区长25 bp,3′非编码区长124 bp,可编码647个氨基酸.生物信息学分析表明,在IAP的N端无氨基酸残基组成的信号肽;与斑马鱼的同源性最好,其次是斑猫鲿;在系统进化上,与斑马鱼、斑猫鲿共聚为1支.用半定量RT-PCR分析正常及细菌诱导下草鱼细胞凋亡抑制蛋白基因在不同组织中的表达分布,正常情况下,凋亡抑制蛋白基因在所取的7种组织中均有表达,其中脑、肠和肾表达最高,肝和心脏次之,鳃和肌肉最低,但差异不显著;用嗜水气单胞菌人工感染24 h后,所有组织中的IAP mRNA水平平均降低约26%,以肝脏和肌肉降低较为明显,在感染48 h后,所有组织中的IAP mRNA水平平均升高约4%,其中心脏、肝脏和鳃分别升高8%、16%、19%,这表明草鱼细胞凋亡抑制蛋白可能参与了机体对嗜水气单胞菌感染的免疫应答. |
