白斑狗鱼雌、雄基因组混池重测序研究

摘    要:为研究白斑狗鱼(Esox lucius)雌、雄基因组差异,实验对20尾雌鱼和20尾雄鱼分别进行混池重测序。测序结果显示:雌、雄池待分析测序数(clean reads)分别为282400688条和277391000条,碱基测序错误率(Q30)为94.44%和93.98%。经过与参考基因组比对分析,得到2076647个雌性SNP和2076942个雄性SNP;105810个雌性Indel和647466个雄性Indel;具有显著性别特异的SNP位点10个;使用SNP-Index算法筛选出性别特异区域26个。对雌、雄基因组进行比对和组装分析,分别获得雌、雄特异序列524条和500条。选择116条雌性和160条雄性特异序列进行PCR验证,得到一条长度为773 bp具有雄性偏向性的序列。

Abstract:Esox lucius,an important characteristic cultured fish in Xinjiang,showing obvious sex-related dimorphism in growth,where the females grow much faster than the males.The mixed pool of 20 females and 20 males E.lucius were sequenced respectively to learn more about the genome difference behind the sex.The sequencing results showed that the number of clean reads in the male pool and the female pool were 282400688 and 277391000,the Q30 were 94.44%and 93.98%,respectively.As a result of sequencing quality conform to the requirements of follow-up bioinformatics analysis,there were SNPs of the male pool and female pool for 2076647 and 2076942.Indel for 105810 and 647466 after mapped with the reference genome,ten sex-specific SNPs were identified,and 26 sex-specific regions were screened by the algorithm of SNP-Index.Sex specific sequences of 524 female and 500 male were obtained by aligned with reference genome and assembled unmapping reads,then randomly selected 116 sequences in 524 female sex-specific sequences and 160 sequences in 500 male sex-specific sequences,and designed specific primers for female and male sex special sequence.One 773 bp male-orientated sequence was screened after PCR verification.These genomic differences in male and female of E.lucius provided a theoretical basis for the study of the sex development mechanism of species and an indispensable technical basis for sex control breeding practice in fish species.
