广西猪乙型脑炎病毒的分离及初步鉴定

摘    要:【目的】了解广西猪乙型脑炎(JE)地方流行毒株特点,为有效防控广西猪JE奠定基础。【方法】对从广西各地猪场采集疑似JE病例猪的脑组织、流产胎儿、死胎、木乃伊胎、公猪精液、猪舍蚊子等靶组织样品进行猪乙型脑炎病毒(JEV)检测,并选择部分阳性样品进行JEV分离鉴定及病毒E基因遗传进化分析。【结果】在采集的465份JE疑似病料样品中,有72份为JEV阳性;通过接种BHK-21细胞、乳鼠及SPF鸡胚,分离获得两株JEV地方分离毒株,分别命名为GP株和LC株;遗传进化分析显示,GP株和LC株之间的核苷酸和氨基酸同源性分别为99.9%和99.8%;两株分离株与疫苗毒株SA14-14-2的核苷酸同源性分别为99.7%和99.9%,氨基酸同源性分别为99.4%和99.6%,且均属GIII型JEV。【结论】广西受检猪群存在较严重的JEV感染,应引起重视并加强防控。

Abstract:【Objective】The present experiment was aimed to understand the epidemics of Japanese encephalitis virus (JEV) in Guangxi, to provide scientific information for its prevention and control. 【Method】The suspected JE disease samples, viz., swine brain, aborted fetus, mummy fetus, boar semen and mosquito samples were collected from pig farms in Guangxi to detect and isolate the JEV, and the phylogenetic analysis of positive isolated strains was conducted based on E genes of JEV strain. 【Result】Out of 465 suspected samples, 72 samples were positive for JEV. After inoculating BHK-21cells in suckling mice and chick embryos, two local strains of JEV were finally isolated and named as LC and GP strains, respectively. The phylogenetic analysis results showed that both, the GP as well as LC strains belonged to genotype Ⅲ and were most closely related to JEV vaccine strain SA14-14-2. GP and LC strains shared 99.7 and 99.9% homology of the nucleotide sequence, 99.4 and 99.6% homology of the amino acid with those of JEV vaccine strain SA14-14-2, respectively. 【Conclusion】The JEV infection was serious in pig farms of Guangxi, and it was suggested to pay more attention to its prevention and strengthening of control measures.
