摘    要:目的]探明低温胁迫下转拟南芥CBF3基因烟草幼苗抗寒性相关生理指标和叶绿体超微结构的变化规律。方法]4℃低温胁迫8 h后,测定转基因烟草幼苗叶片内超氧化物歧化酶(SOD)、过氧化物酶(POD)活性和丙二醛(MDA)含量,同时对叶绿体的超微结构进行观察。结果]低温胁迫转基因烟草幼苗叶片内SOD、POD活性和MDA含量与未转基因烟草之间存在极显著差异(P〈0.01)。透射电镜观察结果表明,低温胁迫转基因烟草叶绿体结构基本是完整的,未转基因烟草叶绿体由杆状变成圆球形,数量有所增加,出现聚集现象,基粒片层混乱。结论]叶绿体的超微结构和生理指标变化结果表明,转基因烟草对低温具有更好的适应能力。

Abstract:Objective]The purpose of this research was to study the changes rules of physiological indices related to cold resistance and chloroplast ultra-structure of transferring CBF3 gene tobacco seedling under low temperature.Method]Under 4 ℃ low temperature for 8 h,the activity of POD and SOD,MDA content and chloroplast ultra-structure of transgenic tobacco seedling leaves were investigated.Result]The activity of POD,the activity of SOD,the content of MDA between transgenic tobacco and normal tobacco plant existed extreme significant deviation(P<0.01).Through transmission electron microscope observation,it was found that the chloroplast structure of transgenic tobacco seedling was elementary complete under low temperature,but the chloroplast structure of wild type had changed from rhabditiform to globular,and its quantity increased,at the same time,they appeared collection phenomenon,grana lamellae unclear.Conclusion]The change results of the chloroplast ultra-structure and physiological indices showed that transgenic tobacco had preferable adaptive capacity for cold temperature.
