摘    要:为获得高质量的冷季型草坪草基因组DNA,用改良和优化常规CTAB提取方法,提取冷季型草坪草基因组DNA,并对所得DNA的质量和浓度进行了检测.结果表明:改良CTAB-B法要好于常规CTAB法和改良CTAB-A法,提取的基因组DNA的OD260/OD280在1.8~1.9之间,电泳条带清晰,RNA消化彻底,DNA无明显降解,其含量和纯度均能满足基因工程操作的要求,且酶切效果理想.

Abstract:Genome DNA was extracted by the improved CTAB method from Cool-season turf grass and the quality and concentration of DNA were detected.The results indicated that improved CTAB-B method was a simple and high efficient method which could obtain the high quality of genome DNA,the value of OD_260/OD_280 was 1.8 to 1.9,the RNA was eliminated completely without obvious degradation of DNA,the content and purity can meet the requirements of genetic engineering,and digested the results were satisfactory.
