摘    要:经过初筛、复筛,从活性污泥中分离出1株富产絮凝剂并适用于甘蔗蔗汁絮凝澄清的微生物菌株A.通过菌株A的5.8S rRNA基因两侧的内转录间隙(ITS)进行序列分析,并结合形态观察确定该菌株为盘多毛孢属(Pestalotiopsis sp).对培养条件进行单因素试验,获得该菌株产絮凝荆的最佳培养条件为:pH值为6.0、28℃、140r/min;培养4d;最佳碳氮源分别为葡萄糖和牛内膏.微生物发酵液在蔗汁沉降试验中的絮凝率为69.2%,清汁色值为87.4IU.

Abstract:By the means of primary screening and double-screening, we obtained a microbial flocculation producing strain A from activated sludge. The strain was identified as Pestalotiopsis sp, by analyzing the sequence of the internal transcribed spacers of the two sides of the 5.8s rRNA gene and the physiologic characteristic and the character of individual form and the colony form of strain with optical microscope and electrical scanning photomicrograph, Which had high flocculation char- acter and could be used in the flocculation and clarification of sugarcane juice. The study carried on the initial optimization experiment test of culture condition of the strain, and obtained the optimal culture conditions under single factor:pH=6.0,4d, 28℃,140r/min, glucose and beef extract respectively used as optimal carbon source and nitrogen source. In the testing of subsidence and clarification decolorization of sugarcane juice,the flocculation rate was 69.2%,and colourity was 87.4IU.
