吉塞拉的组织培养技术研究

摘    要:以大樱桃矮化砧吉塞拉(Gisela)为试材,进行了外植体消毒试验、不同培养时期培养基筛选试验和炼苗移栽试验,并利用植物组织培养技术建立了快繁再生系统。结果表明:吉塞拉组织培养不同培养阶段的适宜培养基分别为:分化培养基:MS+6-BA0.5 mg/L+NAA0.05 mg/L;继代培养基:MS+6-BA0.3 mg/L+NAA0.1 mg/L;生根培养基:1/2 MS+IBA0.3 mg/L。试管苗移栽前先在自然光环境下封闭锻炼然后开瓶炼苗3 d,产生的侧根数量多,移栽成活率高,其中自然光封闭锻炼12 d+开瓶锻炼3 d效果最好。

Abstract:The rapid propagation system of sweet cherry dwarfing stock Gisela was established by tissue culture.The results showed that the suitable differentiation medium,subculture medium and rooting medium was MS+6-BA 0.5 mg/L+NAA 0.05 mg/L,MS+6-BA 0.3 mg/L+NAA 0.1 mg/L,and 1/2 MS+IBA 0.3 mg/L,respectively.The plantlets should be hardened at closed condition with daylight,then opening the glass bottle before transplanting,which would produce more lateral roots,and increase the survival rate of transplanting.The effects of hardening at closed condition with daylight for 12 days and opening the glass bottle for three days were the best.
