摘    要:以综合性状优良的籼稻品种浙恢7954的成熟胚为材料,比较了在遗传转化不同阶段培养基成分对愈伤组织生长及植株再生的影响,进行遗传转化体系的优化。结果表明,最优的愈伤组织诱导培养基为Y1、愈伤组织抗性选择培养基为J3、愈伤组织分化培养基为D6,在此条件下,GFP转化率达26.8%。

Abstract:Agrobacterium meditated-transformations of most rice indica cultivars are far from routine.In this study,the transformation system for an elite indica cultivar,Zhehui 7954,was developed and optimized.The effects of different ingredients in culture media on the growth of callus generated from mature embryo and plant regeneration were investigated.The results showed that Y1,J3 and D6 media were the optimal choice for callus induction,resistance selection and differentiation,respectively.Under the condition,transformation efficiency expressing green fluorescence protein(GFP) was 26.8%.
