摘 要: | 采用甲醛对新城疫LaSota株病毒进行灭活,利用蔗糖透析法进行浓缩,抗原效价由浓缩前的log26上升至浓缩后的log210,在浓缩液中加入25%的丙三醇制成了新城疫血凝抑制抗原.同时对加入稳定剂——丙三醇的自制抗原进行了定质研究.结果表明:加丙三醇自制抗原,温度越低时稳定性越好、保存期最长.将自制抗原、标准抗原与标准血清、待检血清进行血凝抑制试验对比,表明自制抗原准确性、重复性较好,可用于临床监测.

Abstract: | The Newcastle disease virus( LaSota strain) was inactivated by formaldehyde and concentrated through sucrose dialysis method, then the antigen titer increased from log2^6 ( before concentrate) to log2^10 ( after concentrate), after added 25% glycerol as stabilizer to the concentrate virus the hemagglutination inhibition antigen was successfully manufactured, and the antigen was qualitative analysed. The result showed when the temperatare more lower, the self - made antigen more stable and the retention period last more longer. Compare with the standard antigen through hemagglutination inhibi- tion test used standard serum and clinical serum, we found the self - made antigen have good accuracy and repeatability and can be used to clinical survey. |
