摘    要:利用水稻的愈伤组织作受体, 采用PIG基因枪法, 首先成功地将Bt基因cryIA(b)]连同抗除草剂bar基因导入栽培水稻品种中国91, 选育出纯合稳定的株系T91系. 然后将T91系与黄淮区主栽品种豫粳6号杂交, 并辅以标记基因选择, 选育出纯合稳定、综合性状优良的转Bt基因株系C48. 利用农杆菌介导的转化系统将Xa21基因转入C48, 根据标记

Abstract:Using rice calli as explants, a plasmid pBW3 containing cryIA(b) gene and bar gene was transferred into the rice variety Zhongguo 91 with PIG gene gun method, and a line named T91 which was homozygous to cryIA(b) and bar genes was then developed. The cryIA(b) gene was inherited stably into the progenies of the transgenic rice. A rice line C48 which showed ideal agronomic traits was developed by crossing T91 with a cultivated variety Yujing 6 followed by molecular marker-assisted selection. The Xa21 gene, which confers resistance to bacterial blight disease, was then introduced into C48 line via the Agrobacterium-mediated transformation system. Transgenic rice plants containing both cryIA(b) gene and Xa21 gene were obtained. Biological tests showed that the transgenic rice was resistant to both stem borer and bacterial blight disease.
