珙桐产黄酮内生真菌的分离和鉴定

摘    要:采用珙桐(Davidia involucrata Baill.)组织分离产活性物质的内生真菌,以金黄色葡萄球菌、枯草杆菌、大肠杆菌为测试细菌进行内生真菌代谢物对细菌的抑制实验,用高效液相色谱(High Performance Liquid Chromatography,HPLC)分析内生真菌的活性物质成分。结果表明,从珙桐枝、叶中分离到126株内生真菌,其产物对3种细菌的抑菌率分别为65%、71%、25%,其中D37菌株对金黄色葡萄球菌的抑制作用最强。经HPLC分析证明,菌株D37的次生代谢产物中含有槲皮素、山奈酚2种黄酮类物质,每100 g干菌丝可产黄酮31.9 mg。根据菌株D37的形态学特征,结合核糖体基因居间序列(Internaltranscribed spacer,ITS)的分析结果,确定菌株D37为曲霉属,烟曲霉种(Aspergillus fumigatus)。从珙桐分离的产黄酮内生真菌D37可作为生产黄酮类药物的候选菌株,也为抗生素药物的筛选奠定了基础。

Abstract:The endophytic fungi strains producing active substances isolated from Davidia involucrate Baill were tested by Staphylococcus aureus,Bacillus subtilis and Escherichia coli to investigate the inhibiting effect of the metabolic substances produced by endophytic fungi on the tested bacterium and the metabolic substances produced by endophytic fungi were determinated by HPLC.The results show that 126 endophytic fungi strains are isolated from the stems and leavies of Davidia involucrate Baill and the inhibiting bacteria rate of the metabolic substances to Staphylococcus aureus,Bacillus subtilis and Escherichia coli is 65%,71% and 25% respectively.The inhibiting effect of the endophytic fungi strain D37 to Staphylococcus aureus is the highest in all isolated strains.The second metabolic substances of the strain D37 has flavonoids such as quercetin and kaempferol and the strain D37 can produce 31.9 mg flavonoids/100g dry mycelium.The strain D37 is classified as Aspergillus fumigatus according to the morphological characteristic and the similarity of nucleotide sequence of Internal transcribed spacer(ITS) between rDNAs.The strain D37 can be selected as a candidator strain for producing the antibiotic medicine with flavonoids.
