香菇编码线粒体中间肽酶le-mip基因及其紧密连锁基因的克隆

摘    要:根据香菇(Lentinula edodes)编码线粒体中间肽酶le-mip基因的保守序列,通过基因组步行法对le-mip基因及其上游、下游区域进行步行扩增,扩增产物经SeqMan程序拼接后再BlastX搜索,并进行同源性分析.所获得的序列长8 880 bp,其中有5个推定的基因,且与le-mip紧密连锁的基因中没有香菇的A交配型基因.

Abstract:Based on a previously isolated conserved mip gene fragment, chromosome walking was used to amplify the mitochondrial intermediate peptidase gene (mip) in Lentinula edodes, together with the upstream and downstream genomic sequences. Amplified products were connected by SeqMan software, subjected to a BlastX search, and aligned. The total length of combined sequences was 8 880 bp, which together comprised four complete and one partial gene sequences. The A mating type gene in L. edodes is not linked to le-mip.
